A 2-year-old East Friesian sheep imported from Australia exhibited severe anemia after contagious pustular dermatitis in Hokkaido, Japan. Hemoplasma infection was confirmed in blood smears. Both Mycoplasma ovis and ‘Candidatus Mycoplasma haemovis’ were detected by PCR and sequence analyses. In the epidemiological analysis, dual pathogens were detected in 6 of 12 (50.0%) sheep imported from Australia with the infected ewe at the same time, 1 of 5 (20.0%) sheep introduced from a domestic farm in Hokkaido, and in 1 of 16 (6.3%) sheep from an epidemiologically unrelated ranch. It is the first clinical case of sheep to confirm coinfection of these pathogens in Japan. |

To better understand the mechanism of excessive gas accumulation in the abomasum in bovine abomasal displacement, we performed gastric fluoroscopy in vagotomized cattle. Fifteen 6-month-old Holstein steers were divided into three groups: a non-vagotomized control group (Group C; n=5), a ventral thoraco-vagotomized group (Group V; n=5), and a dorsal and ventral thoraco-vagotomized group (Group DV; n=5). These groups were examined by fluoroscopy before and during a 5-week observation period after surgery. In Group C, no change was observed throughout the observation period. Babesia ovata is a tick-transmitted hemoprotozoan parasite that infects cattle. In our study, bovine blood samples (n=2,034) were collected from 10 different countries (Brazil, China, Ghana, Japan, Mongolia, the Philippines, South Africa, Sri Lanka, Thailand and Vietnam) and DNA extracted. The DNA samples were screened using an established and specific polymerase chain reaction (PCR) assay targeting the Apical membrane antigen 1 (AMA-1) gene. Parasite DNA was detected among samples collected from Japan, Mongolia and Thailand. Two dogs that exhibited sudden astasia, anorexia and fever higher than 40°C were suspected of having Lyme disease in July 2011. Clinical symptoms gradually improved with antibiotic treatment in both cases. Polymerase chain reaction and sequence analysis revealed Borrelia garinii DNA fragments in the peripheral blood in the acute disease phase. Serological tests, including enzyme linked immunosorbent assay and Western blot analysis, showed an increased IgG antibody titer against Borrelia pathogens in one of the dogs. These findings suggested that diagnosis of the two dogs was Lyme disease related to B. garinii infection. |

Computed tomography (CT) was used for diagnosis of brain abscess in a 6-month-old, Japanese black calf presented with neurological dysfunction, compulsive circling and vision disturbance. CT images showed asymmetric lateral ventricles, and presence of intra-cranial multiple low absorption lesions surrounded by capsule suggestive of abscess in the right cerebral hemisphere. Postmortem examination revealed marked swelling of right cerebral hemisphere and olfactory bulb. Multilocular large abscess containing creamy pus was found to occupy most area of periventricular and lateral ventricle. A 12-month-old Holstein heifer with anorexia, lameness, and enlargement of peripheral lymph nodes was suspected of having bovine leukosis. Although lymphocytosis was not observed, cytology of fine needle aspirate from a superficial cervical node, and increased serum lactate dehydrogenase and thymidine kinase activities, strongly suggested lymphosarcoma. Increased numbers of mononuclear cells as well as mitotic cells were observed in synovial fluid collected from swollen joints. Pathological examination confirmed B-cell calf form bovine leukosis and joint swelling related to neoplastic cell infiltration. 黒毛和種牛が出生翌日より横臥,後弓反張を呈した.脳挫傷を疑い,抗生物質,ビタミンB1製剤,消炎鎮痛剤により加療され,いったんは改善したが同様の症状が間欠的に認められた.歩様異常は継続して認められたが,一般状態は良好で8カ月齢まで飼養された.しかし,その後開脚姿勢,測定過大を伴う歩様異常,威嚇瞬き反応の低下が進行した.病理組織学的検索により小脳にプルキンエ細胞の顕著な変性と脱落及び顆粒細胞の減数を認め,小脳皮質変性症と診断された. |

38カ月齢のホルスタイン種乳牛において,体全体が左側に傾くという神経症状が進行性に認められた.経過,左側対光反射と左側音響耳介反応の減弱,及び聴性脳幹誘発電位測定検査における左刺激時Ⅲ波-Ⅴ波の遅延所見より,延髄から橋における片側性圧迫性病変を疑った.病理解剖の結果,橋左側に帯黄色脆弱化病巣を認め,病理組織学的検索により,病変は腫瘍であり,迷入組織に由来する可能性があるものの,原発組織は確定できなかった. |

The clinical usefulness of antibodies against Babesia gibsoni detected by ELISA with recombinant P50 was examined in dogs in an area where B. gibsoni infection was endemic. Only 8 among 14 dogs with acute type B. gibsoni infection without a previous history of infection were positive. This high percentage of false-negative results is thought to be a weak point of ELISA as a diagnostic tool. However, 14 other anemic dogs with a confirmed history of B. gibsoni infection were all positive, thus confirming the higher sensitivity of ELISA in detecting a history of infection. |

We investigated the concentration of the bacterial endotoxin lipopolysaccharide (LPS) in the blood, ovarian follicular fluid and uterine fluid of a clinical case of bovine metritis. A 2-year-old lactating Holstein cow exhibited continuous fever >39.5°C for more than 2 weeks after normal calving. The cow produced a fetid, watery, red-brown uterine discharge from the vagina and was diagnosed with metritis. The LPS concentrations in plasma and uterine fluid were 0.94 and 6.34 endotoxin units (EU)/ml, respectively. One of seven follicles showed an extremely high level of LPS (12.40 EU/ml) compared to the other follicles (0.62–0.97 EU/ml). These results might suggest the presence of high concentration of LPS in follicles in cows with postpartum metritis. |

The prevalence of Ehrlichia and Anaplasma in 1,427 dogs from 32 Japanese prefectures was evaluated by PCR and DNA nucleotide sequencing. PCR screening demonstrated that 18 dogs (1.3%) were positive for Anaplasmataceae. Sequence analysis revealed that 14 of the amplicons were most closely related to Wolbachia spp., symbionts of Dirofilaria immitis, whereas three were identified as Anaplasma bovis. The remaining amplicon could not be sequenced. Babesia and Hepatozoon infections of dogs in a village of eastern Sudan were analyzed by using a single PCR and sequencing. Among 78 dogs, 5 were infected with Babesia canis rossi and 2 others were infected with B. canis vogeli. Thirty-three dogs were positive for Hepatozoon. Hepatozoon canis was detected by sequence analysis. |

DNA fragments of 'Candidatus Mycoplasma haemominutum', a feline heamobartonella pathogen, were detected from unfed Ixodes ovatus collected from vegetation in Hokkaido, Fukushima and Yamaguchi Prefectures, and unfed Haemaphysalis flava in Yamaguchi Prefecture. This finding suggests that ixodid tick is a possible vector of 'C. Mycoplasma haemominutum'. Spiroplasma DNA was also detected from unfed I. ovatus in Hokkaido, Fukushima and Yamaguchi Prefectures. The analysis of nucleotides sequence suggested that this Spiroplasma was distinct from registered species. |

DNA from ticks recovered from 1137 dogs and 133 cats from Ell over Japan were examined for Rickettsia infection by citrate synthase gene (gltA)-based PCR and partial nucleotide sequencing. A total of 91 dog tick samples and 18 cat tick samples showed a single band of the appropriate size in the nested PCR. Sequence analysis was successfully performed on 102 samples. DNA of Rickettsia japonica or closely related Rickettsia spp. strains were detected from 38 licks in 16 prefectures mainly in western Japan.
